The control group for MAO activity included 95 men mean ageSD 4213 years ; and 43 women mean age 3512 years ; . Women displayed a significantly higher enzyme activity than men, as the mean platelet MAO activity was 12.45.3 nmol mg protein h range 3.125.8 ; in the first group, and 9.05.2 nmol mg protein h range 1.931.9 ; in the latter group. The mean value of lymphocyte MR binding did not significantly differ between males n 86; mean age 4310 years ; and females n 69; mean age 3710 ; . In men it averaged 12.29.9 fmol 106 cells range 1.037.9 ; and in women 10.79.7 fmol 106 cells range 1.139.7 ; . Altogether these data indicate i ; wide inter-individual differences in the values of platelet MAO activity and lymphocyte MR binding in healthy subjects; ii ; gender-related differences in MAO activity, but not in MR binding. The application of these assays to a cohort of 7 year-old Faroese children exposed to methylmercury and PCBs through the diet has been recently undertaken to assess whether these peripheral markers are altered by environmental exposure to these neurotoxicants. Chronic pain is a common neurological problem that is largely intractable to current therapies, despite improvements in the understanding of mechanisms contributing to pain following tissue and nerve injury. This study investigates a role for integrins in mediating such pain. Integrins are adhesion molecules that bind extracellular matrix ECM ; proteins, including laminin and fibronectin. They are transmembrane heterodimers comprised of and subunits ; that mediate signalling in both directions across the membrane. They are present on primary afferent nociceptors and are thus optimally located to sense changes in the ECM environment, which accompany inflammation and injury. In this study, several strategies were used to disrupt integrin signalling, which concomitantly inhibited the development of hyperalgesia in rat models. Peptide fragments of laminin or monoclonal antibodies against the 1 and 3 integrin subunits involved in laminin binding ; blocked the hyperalgesia triggered by the injection of prostaglandin E2 PGE2 ; , and carageenan a longer-lasting model of inflammatory pain ; , but not by adrenaline. Fibronectin peptides and antibodies against the 5 subunit of integrin involved in fibronectin binding ; blocked hyperalgesia induced by adrenaline, but not by PGE2 or carageenan. The 1 subunit is involved in both laminin and fibronectin binding. Monoclonal antibodies and antisense deoxynucleotides against this subunit prevented hyperalgesia induced by all the above triggers. The 1 subunit antisense molecules also inhibited taxolmediated hyperalgesia, which is a model of neuropathic pain. Thus, it appears that integrins in fact mediate pain through several pathways. This demonstration that integrins play a significant role in cell signalling pathways mediating sensitisation of nociceptors, not only adds to our knowledge of chronic pain mechanisms, but also offers hope for the development of better therapies for those suffering from inflammatory and neuropathic pain. -LMS, SJT. The effects of posterior internal pallidal ablation GPi pallidotomy ; on parkinsonian signs and symptoms were studied in 15 patients with medically intractable Parkinson's disease PD ; . The sensorimotor territory of the internal portion of the globus pallidus and the adjacent optic tract and internal capsule were identified with microelectrode recording and stimulation. Radiofrequency lesions were then created in the identified sensorimotor territory. Pallidotomy significantly improved all cardinal parkinsonian motor signs tremor, rigidity, akinesia bradykinesia, and gait dysfunction ; and reduced drug-induced motor fluctuations and dyskinesias. The improvements occurred predominately contralateral to the lesion, but were also present ipsilaterally. Early postoperative 3-month ; , mean total United Parkinson's Disease Rating Scale scores improved by 30.1% from preoperative values. Mean combined "on off" Schwab and England Scale scores, a measure of functional independence, increased from 48.8% to 73.0% postoperatively. The mean total United Parkinson's Disease Rating Scale and Schwab and England scores did not show a statistically significant decline over the 1-year postoperative period. Surgery resulted in little morbidity, including a lack of significant deficits on neuropsychological and psychiatric testing. Physical and social functioning and vitality measures on the Medical Outcome Scale also showed significant improvement over the postoperative period. The findings of this pilot study demonstrate that ablation of the sensorimotor portion of the internal pallidum is a highly effective treatment for advanced PD, with benefits sustained at 1 year.

There have been a number of changes to US patent law over the last 6 years in my opinion, more than in the previous 60 years total. These changes include changes in patent term, changes in public access to files prior to issuance of patents, and changes in ability to enforce patents. These changes have had significant effects on how companies use their intellectual property, especially with respect to patent term, and how US patents can be used in "industrial espionage". For example, to claim a gene encoding a particular protein, one must provide the sequence of the gene to comply with the written description requirement, although it may be possible to enable someone to obtain and use the gene merely by providing a source and appropriate primers. Although legally there is no requirement for "actual reduction to practice" as of the date the application is filed, in practice we are finding that applications are being routinely rejected as "non-enabled" if there is no actual data in the application, at least in fields that are allegedly "unpredictable". Unpredictable areas include tissue engineering, gene therapy and treatment of diseases such as cancer and AIDS. We also see rejections based on data obtained in vitro on the grounds that it is allegedly not predictive of results obtained in vivo. Accordingly, particularly in a field where there is a lot of "constructive reduction to practice" i.e., where many patent applications describe what could be done, rather than what has actually been done ; , we recommend including any data possible, or literature support, in the application, in anticipation of these rejections. The test in all of these cases should be whether those skilled in the art believe the description to be adequate to lead those skilled in the art to what is being claimed, without undue experimentation, with a reasonable expectation of success not absolute certainty ; . The law has always been that one does not need to have actually made an invention in order to file a patent application claiming the invention, if the specification tells one of ordinary skill in the art as of the date of filing of the application how to make and use that which is claimed. Of course, the key is what is known at the time of filing and what is the level of skill in the art. I recently had a patent interference, a litigation type proceeding in the USPTO to determine priority of invention between multiple parties, which is then determinative of which of the parties who are claiming the same invention is entitled to the patent. The entire outcome was determined based on a question of what one of skill in the art could have done with only routine experimentation at the time of filing of the first application, compared with what one of skill in the art could have done with only routine experimentation as of the date of the second application. Even though the second party had done exactly the same things that the first party had said to do but not actually done ; , the first party lost the interference based on essentially ; attorney argument and literature references casting doubt on the ability of the person of ordinary skill in the art, as of the date of the first filing to practice all of the claimed embodiments. Depression is a common emotional consequence in MS.
